Resumen:
La enfermedad de Alzheimer (EA) es la demencia degenerativa primaria más
frecuente, con un incremento de lapoblación de riesgo en la última
década. El deterioro cognitivo (DCL) leve es un cuadro limítrofe
entre la normalidad y laEA, presentando una taza muy de conversión a
esta enfermedad 12 al 15% anual con un 50% a los 5 años. La búsquedade
marcadores que permitan predecir la conversión de DCL a EA y el declive
cognitivo en la EA leve es un área relevantede la investigación
Mejores biomarcadores que evalúan la presencia de EA, pero no su pronóstico
evaluando βamiloide y la Punción Lumbar pero estos son costos
y traumáticos. Los estudios actuales coinciden que es necesarioel desarrollo
de nuevos marcadores cognitivos y biomarcadores que superen estas limitaciones.Recientemente,
se desarrolló un modelo transgénico en ratas que recapitula etapas
tempranas de la EA. Reportamos alteracionesbioenergéticas en sinaptosomas
aislados de hipocampo asociadas a déficits cognitivos. Se ha postulado
al estadobioenergético como potencial biomarcador, Proponemos como hipótesis
de trabajo que la memoria episódica (ME)junto a la memoria lógica
(ML) es el principal marcador cognitivo y que la funcionalidad bioenergética
plaquetariaconstituiría un posible biomarcador periférico de progresión
del deterioro cognitivo en EA. Los objetivos son: Evaluarsi la ME y ML funcionan
como marcador cognitivo. Determinar si el BHI plaquetario predice la progresión
del deteriorocognitivo es predice del empeoramiento en pruebas cognitivas en
el modelo transgénico. Evaluar la relación entremarcadores cognitivos
y bioenergeticos Se reclutarán personas ≥ 65 años sanas,
con DCL o con EA leve. A lospacientes se les realizara, evaluación neuropsicológica
marcadores de severidad MMSE y GDS y en ambos determinarla memoria verbal, la
memoria lógica el BHI plaquetario. Se repetirá el mismo procedimiento
transcurridos 1 y 2 años.Simultáneamente, se determinará
el BHI plaquetario y el desempeño en tareas hipocampo-dependientes en
ratas wild-type y transgénicas hemicigotas a los 3 y 12 meses de edad.
En ambos estudios, se realizarán análisis estadísticos
a finde determinar si el BHI plaquetario es predictor de la progresión.
Si se confirmara nuestra hipótesis, el BHI plaquetariosería un
biomarcador de progresión, que junto con la memoria podrían alcanzar
un alto valor predictivo

COGNITIVE AND PLATELET MARKERS IN THE EVOLUTIONARY PROGNOSIS OF ALZHEIMER'S DISEASE AND MILD COGNITIVE IMPAIRMENT.
Alzheimer's disease (AD) is the most common primary degenerative dementia,
with an increased risk population inthe last decade. Mild cognitive impairment
(DCI) is a borderline between normality and AD, with a very high rate ofconversion
to this disease 12 to 15% annually with 50% at 5 years. The search for markers
that predict the conversionof DCL to AD and cognitive decline in mild AD is

in AD. The objectives are: To evaluate if the ME and MLfunction as a cognitive
marker. Determining whether platelet BHI predicts progression of cognitive impairment
ispredictive of worsening cognitive testing in the transgenic model. To evaluate
the relationship between cognitive andbioenergetic markersPeople aged ≥
65 years will be recruited, with DCL or with mild AE. Patients will be assessed,neuropsychological
markers of severity MMSE and GDS and in both determine the verbal memory, the
logical memorythe platelet BHI. The same procedure will be repeated after 1
