Análisis comparativo-evolutivo de las bases neurales del aprendizaje: Estudio funcional de la Cognición espacial, Expectativa y Frustración.

Resumen:
El objetivo del presente proyecto es estudiar con un abordaje comparativo-evolutivo
los mecanismos que subyacen a la cognición espacial y a los fenómenos
conductuales asociados a cambios sorpresivos de una recompensa (relacionados con
la respuesta de frustración). Usando modelos animales (anfibios y ratas),
se estudiará el papel de la memoria de los reforzadores previos y su modulación
por variación de factores ambientales y por la administración de
fármacos agonistas y antagonistas de distintos sistemas de neurotransmisión.
Además, analizaremos la actividad neural en distintas áreas del
cerebro a través de técnicas neuroinmunohistológicas. En
sapos, a partir de trabajos previos desarrollados en nuestro laboratorio, se estudiará
el aprendizaje de geometría espacial y claves de referencia, analizando
el papel del pallium medial (la formación hipocampal de los anfibios).
Por otra parte, también se continuará con el análisis del
aprendizaje discriminativo de soluciones salinas en anfibios. Además, indagando
acerca de su origen evolutivo, se estudiará la posible existencia del efecto
de contraste negativo sucesivo consumatorio en sapos, usando distintas soluciones
salinas como reforzadores apetitivos. En ratas, se caracterizará una situación
de contraste negativo sucesivo instrumental, realizando también registros
neuroinmunohistológicos de la actividad cerebral asociada. Por último,
se analizará el efecto de la administración de fármacos opioides
y de moduladores de la memoria sobre el contraste negativo instrumental. Globalmente,
se espera: (1) obtener información para una mejor comprensión de
la relación entre procesos cerebrales y conducta; (2) determinar cuán
conservados están estos fenómenos en la filogenia, y (3) describir
los circuitos cerebrales básicos que subyacen a estas conductas, conocimiento
relevante para desarrollar potenciales terapias de recuperación de funciones
cognitivas en pacientes con deterioro cerebral que afecte a ese tipo de conductas.

The goal of this project is to study with a comparative-evolutionary approach
the mechanisms underlying spatial cognition and behavioral phenomena associated
with surprising changes of a reward (related to the frustration response). Using
animal models (amphibians and rats), the role of the memory of the previous reinforcers
and their modulation by variation of environmental factors and by the administration
of agonist and antagonist drugs of different neurotransmission systems will be
studied. In addition, we will analyze neural activity in different areas of the
brain through neuroimmunohistological techniques. In toads, taking into account
previous work developed in our laboratory, we will study the learning of spatial
geometry and reference cues, analyzing the role of the medial pallium (the hippocampal
formation of amphibians). On the other hand, we will also continue with the analysis
of discriminative learning of saline solutions in amphibians. In addition, inquiring
about its evolutionary origin, we will study the possible existence of the consummatory
successive negative contrast effect in toads, using different saline solutions
as appetitive reinforcers. In rats, a situation of instrumental successive negative
contrast will be characterized, also making neuroimmunohistological analysis of
the associated brain activity. Finally, we will analyze the effect of the administration
of opioid drugs and memory modulators on the instrumental negative contrast. Overall,
it is expected: (1) to obtain information for a better understanding of the relationship
between brain processes and behavior; (2) to determine how preserved these phenomena
are in phylogeny, and (3) to describe the basic brain circuits that underlie these
behaviors, relevant knowledge to develop potential therapies for the recovery
of cognitive functions in patients with brain deterioration affecting this type
of behaviors.
